5-ht2 Receptor
|
The 5-HT2 receptors are a subfamily of 5-HT receptors that bind the endogenous neurotransmitter serotonin (5-hydroxtryptamine, 5-HT). The 5-HT2 subfamily consists of three G protein-coupled receptors (GPCRs) which are coupled to Gq/G11 and mediate excitatory neurotransmission, including 5-HT2A, 5-HT2B, and 5-HT2C. Is the efficacy of SSRI's among those with premature ejaculation sufficient evidence that this condition may be etiologically related to dompaminergic dysfunction? Or, do these drugs simply over-ride what may best be dealt with physically or psychologically, perhaps via masturbating regularly or before sexual engagements in combination with some cognitive-behavioural exercise. One example from the literture supporting a dopamine hypothesis [I probably could have found something better]: Just some background: SSRI's can induce reduction in dopamine, delaying orgasm--it is well known that dopamine is the key neurotransmitter in stimulating orgasm. Essentially, SSRI's *can* work by stimulating postsynaptic 5-HT2 and 5-HT3 receptors, reducing… [cont.] Asked by Kynysca - Tue Dec 9 18:12:28 2008 - Medicine - 2 Answers - Comments A. I can't comment on the ejaculation aspect of your question (well, I guess I could, but I won't), but I would like to ask about your comment that SSRIs might reduce DA release in the substantia nigra. This region is an area of dopaminergic soma, and unless we talk of dendritic release, I'm not sure these drugs will affect DA release in SN to any significant extent. Perhaps you meant release from striatum, nucleus accumbens, etc., or some other brain regions receiving innervation from the substantia nigra. Answered by Simonizer1218 - Wed Dec 10 06:49:23 2008 How does Olanzapine work (Question for pharmacists, or other healthcare profesionals)?
Q. I understand that Olanzapine is an antagonist at both dopamine D2 and 5-HT2 receptors (among others). Now, I can see why Olanzapine may reduce the positive symptoms of schizophrenia and related disorders as it acts to block dopamine at the receptor site, but I can't work out why introducing a 5-HT2 antagonist (as olanzapine is) would reduce the negative symptoms. Surely this would decrease serotonin transmission in the brain and increase the negative. I note that aripiprazole is a 5-HT2 agonist and I can understand why it would reduce negative symptoms. If someone could please explain Olanzapines mode of action, I'd be most grateful. Yes, I too have been on Olanzapine, albeit a low dose (for anxiety/paranoia from traumatic event). I am not a doctor or pharmacist but I was on zyprexa for years and it certainly does increase negative symptoms. I think it is perhaps the worst 2nd generation anti psychotic there is. I took an SSRI with it to help with serotonin but it didn't really help much.Do you have any of the literature that comes with the medication?
